Regioselective alkylation at the 3-position of 2-amino or 2-acylaminopyrazine was achieved with organolithium in THF at -20 oC. Additives such as TMEDA, CuI and BF₃-Et₂O, oxidative workup and metal exchange to MgBr or Cu gave no improvement.
